The Relationship between Polymorphisms of Wnt Antagonists Genes and Production of Osteoprotegerin (OPG)-soluble receptor Activator of NF-¥êB Ligand(sRANKL) by Whole Blood Cells

Abstract
Objectives: To investigate the association between polymorphisms of Wnt antagonists genes and the production of
osteoprotegerin (OPG), and soluble receptor activator of nuclear factor-¥êB ligand (sRANKL) by whole blood cells
(WBCs) in postmenopausal Korean women.
Methods: The Dkk1 c.318A>G, Dkk2 c.437G>A, Dkk3 c.1003A>G polymorphisms and sFRP3 c.970c>G, sFRP4
c.958C>A, and c.1019G>A polymorphisms, and sFRP5 c.20G>C polymorphisms were analyzed by Taqman assay,
DNA sequencing, and restriction fragment length polymorphism (RFLP) in 156 Korean postmenopausal women.
The production of OPG and sRANKL by lipopolysaccharide-stimulated WBCs were also measured.
Results: The production of OPG by lipopolysaccharide-stimulated WBCs was related with the Dkk1 c.318A>G
polymorphism. The AA genotype of the Dkk1 c.318A>G polymorphism showed a higher production (P=0.04) of
OPG by lipopolysaccharide-stimulated WBCs than GG genotype. No significant differences in the production of
OPG and sRANKL by WBCs and their ratio were noted according to single polymorphisms of the sFRP genes and
the haplotype genotypes of sFRP4 c.958C>A and c.1019G>A polymorphisms.
Conclusion: The Dkk1 c.318A>G polymorphism among Dkks and sFRPs polymorphisms measured affects the production
of OPG by lipopolysaccharide-stimulated WBCs
